Maladie de Behçet‎ : recherche de marqueurs biologiques et caractérisation d'anticorps anti-VE-cadhérine

Abstract : Introduction: Behçet's disease (BD) is a chronic multisystemic inflammatory disease. Pathogenesis of BD involves neutrophilic hyperactivity, activated endothelium, humoral and cellular immunity. Aim: To find biological markers for the diagnosis and the follow-up of BD. Materials and Methods: All patients who consulted with ocular BD at the University Hospital of Grenoble, between January 2008 and February 2009, were included. Clinical data and blood samples were gathered. The following biological parameters were investigated: circulating VE-cadherin, anti-VE-cadherin antibodies, anti-HSP70 antibodies, plasmatic elastase level, myeloperoxidase level, adenosin deaminase plasmatic activity, neutrophilic NADPH oxidase activity and TNFα and IL-6 supernatant concentrations released by non-CD19 peripheral blood mononuclear cells after stimulation. Results: Anti-VE-cadherin antibodies, anti-HSP70 antibodies and plasmatic elastase levels were increased in BD compared to healthy people (Respectively p = 0.00021, p < 0.001, p = 0.03). None of these biological markers was correlated with general or ocular activities of BD. From a neuro-BD patient's serum with a very high anti-VE-cadherin antibodies level, we showed that anti-VE-cadherin antibodies were aimed against the VE-cadherin extracellular domains 3-4. Conclusion: We described for the first time anti-VE-cadherin antibodies in BD.
