Étude préliminaire de deux composés thérapeutiques potentiels pour le traitement du follicule pileux dans l'Hidradénite suppurée

Abstract : Verneuil's disease or Hidradenitis suppurativa (HS) is a disease of the hair follicle which causes skin inflammation. The hypothesis of an immune deficiency towards skin commensal flora in patients with a genetic susceptibility seems now confirmed. The study of the response of outer root sheath keratinocytes of patient’s hair follicles against Pathogen Associated Molecular Patterns by their Pattern Recognition Receptors demonstrated a decreased production of antimicrobial peptides as psoriasin compared to healthy individuals. Moreover, genetic studies performed on patients have revealed the presence of mutations in the γ-secretase, a key enzyme in the Notch signaling pathway. The team has demonstrated that this signal defect may induce a decreased production of antimicrobial peptides after stimulation of cells PRRs. The aim was to find a compound to restore the NOTCH signal in HS patients, in order to restore the production of antimicrobial peptides by keratinocytes. The study on HaCaT and HEK293T cell lines with two compounds A and B showed that compound A could restore NOTCH pathway but not compound B. Finally, preliminary results on primary cells from healthy individuals indicate that the compound A inhibits antimicrobial peptides production whereas the compound the compound B does not.
