Recherche de partenaires de Lu/BCAM dans le rein chez la souris Lu+/+ et Lu-/-

Abstract : The glycoproteins (gps) Lutheran (Lu) and BCAM (Basal Cell Adhesion Molecule), are adhesion molecules and receptors to laminin 511/521. Both isoforms are encoded by a single gene LU. They belong to the Superfamily of Immunoglobulins (SFIg) and the two isoforms differ only by the size of their cytoplasmic domain. The Lu isoform has 40 additional amino acids compared to Lu(v13) isoform, among which we note the presence of several phosphorylation sites. Lutheran human glycoproteins are expressed in red blood cells, in vascular endothelial and in many tissues as heart, skin, intestine, kidney... namely on epithelial, endothelial and smooth muscle cells. In mice, the Lu protein is expressed in many tissues (heart, lung, kidney, intestine, muscle ...), but it is totally absent on red blood cells. The team with which I did my internship did the inactivation of the gene in mice. Lu/BCAM-/- mice develop normally but show structural abnormalities of the basement membrane in particularly kidney and intestine. Lu participates in maintaining the integrity of the glomerular basement membrane and basal lamina surrounding the intestinal smooth muscle cells. Lu/BCAM is thus essential to the cohesion of the kidney tissue and intestinal muscle. My project concerns the research of the Lu/BCAM partners potentially involved in maintaining the integrity of the filtration barrier (transmembrane partners, components cytoskeleton, proteins involved in intracellular signaling …). I participated to the development of the technical co-precipitating, and then the protocol identification of potential candidates by mass spectrometry. (Platform Jacques Monod, University Paris Diderot). The technique was first tested trying to co-precipitate a known partner protein of the Lu/BCAM, the α5 laminin chain. The results are not successful. The technicale development is in progress.
