La transmission du signal de la cohésion au centromère du chromosome métaphasique

Abstract : Cohesin complex maintains cohesion along chromosomes’ arms and at centromere during the first stage of prophase. The phosphorylation of cohesin subunit SCC2 by Plk1 and Aurora B leads to a loss of cohesion on chromosomes’ arms during the prophase pathway. At centromere, some proteins protect cohesin complex of this pathway until the metaphase. The aim of the project described in this report is to define the transduction of cohesion signal at centromere on the metaphase chromosome. In this study, we used siRNA knockdown on proteins of interest and we shown the impact of this depletion using immunofluorescence on the other proteins of interest. We also see the involvement of CENP A (a centromere specific variant of histone H3) serine 7 phosphorylation by Aurora A and Aurora B in the support of cohesion at centromere. We first identified the centromeric localization of Aurora A by immunoflurescence and, in a second time, we used CENP A Serine7 mutated to alanine cells lines and observed a loss of cohesion. At the end of the study we can certainly give the first steps of the pathway. First, HDAC3 allows BUB1 targeting at centromere. BUB 1 initiates two ways: the first one, involving Aurora A and CENP A S7 phosphorylation leads to Aurora B localization at centromere. The second way leads to Shugoshin 1 localization at centromere. The relationship between Aurora B, Shugoshin 1 and Haspin was not clearly identified at the end of this internship.
