Le rôle des phospholipases A2 sécrétées dans la fertilité : action sur la réaction acrosomique des spermatozoïdes humains

Abstract : Recent studies on mice show the important role of secreted phospholipases A2 (sPLA2) during capacitation and acrosome reaction, two crucial phenomena occurring during fertilization, neglected so far in human fertility. It also has been shown that treatment of mice sperm with mGX (mouse Group X sPLA2) improve these phenomena and results of Assisted Reproductive Technics (ART). The main goal of this study was to evaluate the RA triggering of human sperm by sPLA2. We tested hGX, hGV, hGIb (human sPLA2) and mGX. We also compared the effect of mGX and an inactive mGX in order to determine the effect of catalytic activity on RA triggering. Thus, we tried to characterize the response of patients to sPLA2 with their clinic and biologic characteristics. For each patient, we realized: - RA assessment with or without sPLA2; - an hormonal and lipids check-up; - semen parameters assessment. 61 patients have been included in Grenoble center of ART-CECOS. Our data show the significant RA-triggering by hGV (p<0.001), hGX (p<0.001) and mGX (p=0.001). The inactive mGX has no effect on RA which lead us to the conclusion that sPLA2 act on RA by their catalytic activity. Then, it seems that the response to sPLA2 is independent to semen parameters, age, type of infertility, BMI or lipid status. Our study shows the role of sPLA2 on human sperm. These data and animal models invite us to keep up the investigations on sPLA2 in human fertility in order to develop the sPLA2 use on ARTs.
