Métabolisme des lipoprotéines et biomarqueurs dans les maladies cardiovasculaires

Abstract : Despite the huge progress made in medicine, cardiovascular diseases remain one of the greatest causes of death in the industrialized world. The discovery of novel biomarkers to evaluate treatment’s efficacy at the preclinical and clinical levels is a major stake for the pharmaceutical industry. The first project in this engineering memory involves the measurement of pre-beta HDL concentrations. This first study clearly demonstrated that pre-beta HDL is highly unstable in plasma and that its concentration is dependent on using inhibitor of the enzymes LCAT and LPL. The second concerns the measurement of plasma microparticles using flow cytometry in patients with familial hypercholesterolemia undergoing apheresis. The results have shown that apheresis can decrease total microparticle concentrations in plasma. The third project involves the measurement of CRP kinetics using mass spectrometry in obese volunteers before and after weight loss. In this study, I demonstrated that it is possible to measure the turnover of CRP in plasma using stable isotopes and mass spectrometry; the results suggest that weight loss decreases the hepatic output of CRP. Collectively, these results have proved informative, further work is required.
