Progression tumorale et résistance à la chimiothérapie induites par la stimulation de TLR7 dans le cancer du poumon non à petites cellules

Abstract : 70% of non-small cell lung carcinoma patients have tumoral cells positive for TLR7, a receptor in innate immunity. On in vitro experiments with cell lines, TLR7 induces an increase in tumor cell growth and resistance to chemotherapy. We studied in vivo the effects of TLR7 stimulation on tumor cell lines and the expression of TLR7 on a non-small cell lung carcinoma cohort of patients. In two immunodeficients murine models, TLR7 stimulation induces an increase in tumor cell growth and resistance to chemotherapy. In an immunocompetent murine model, TLR7 stimulation can lead both to tumor decrease through immune cells stimulation or to an increase in tumor cell growth and resistance to chemotherapy, depending on TLR7 agonist dose. In the non-small cell lung carcinoma cohort of patients, a strong expression of TLR7 on tumor cells is correlated to a bad prognosis survival. These results show that TLR7 can possibly be used as a predictive marker of patient’s response to treatment and other studies might be conduct to study the possible adaptation of treatment according to TLR7 status.
