Purification et caractérisation biologique de polyphénols polyprénylés issus de Garcinia bancana capables de moduler l'expression de molécules du Complexe Majeur d'Histocompatibilité (CMH)

Abstract : In organs transplantation, current immunosuppressive treatments are used to control the immune response directed against the graft but they have important secondary effects. The search for new therapeutic strategies is then essential. Previous works within our team have shown that a polyprenylated polyphenol, guttiferone J, was able to modulate the expression of molecules involved in the immune response (molecules of the Major Histocompatibility Complex (MHC)). Thereby, this work, first of all, consisted in the purification by semi-preparative HPLC of some new polyprenylated polyphenols extracted from Garcinia bancana and their identification: xanthochymol and guttiferone F which, by structural analogy, may have effects similar to guttiférone J. Then, in a biological study by flow cytometry, qPCR and Western Blot, a decrease in the expression of MHC molecules, especially HLA-E, was observed with these compounds. This resulted in a decrease in surface protein, a decrease in the expression and phosphorylation of the transcription factor STAT1 involved in the signaling pathway of IFN-γ, activated during immune responses. Thus, these natural products constitute new STAT1 inhibitors whose inhibition mechanisms remain to be further investigated.
