Utilisation d'agents chimiques pour l'amélioration des phases post-fécondation chez la perche européenne Perca fluviatilis

Abstract : Fish eggs have interspecific particularities that can be problematical during a few farming phases. In European perch Perca fluviatilis eggs are surrounded by a glycoproteic matrix tying them together and forming a long ribbon. The latter is a source of complication in hatcheries during incubation or hatching. The aim of this study is to find a reagent reducing those complications by affecting the ribbon or the eggs’ structures. Work has been divided in two parts, using for each of them ribbons collected from the Valperca SA’s hatchery. On the one hand, we tried to dissolve the ribbon after artificial fertilization in order to obtain free eggs. On the other hand, we tried to synchronize hatchings by dissolving the ribbon and the eggs membrane. The following reagents were chosen from existing bibliography and tested: tannic acid, Alcalase®, protease from bovine pancreas, sodium hypochlorite, bromelain and papain. Tests of the first part were not conclusive even though papain seems to be the able to modify the ribbon structure. Further researches are needed. However, synchronization tests gave interesting results. Alcalase® was very efficient, giving a hatching rate higher than 75% 3 hours after the chemical reaction, increasing in the same time the number of larvae obtained. Those results are encouraging for the firm but bigger tests still need to be done.
