Troubles métaboliques et risques cardiovasculaires associés à la prise d'antipsychotiques de deuxième génération‎

Abstract : Second-generation antipsychotics are recommended as first-line treatment for psychiatric disorders such as schizophrenia. They are, therefore, more and more used. Practitioners are therefore confronted with metabolic side effects and cardiovascular risks when taking these treatments by their patients. The aim of this thesis is, in a first part, to present what antipsychotics are: their history, their therapeutic effects, their indications, their classifications according to their clinical effects or their chemical structure, and their mechanisms of action. A second part presents their metabolic and cardiovascular side effects (obesity, dyslipidemia, type 2 diabetes, metabolic syndrome, QT prolongation). For each adverse effect, its pathophysiology as well as the impact of second-generation antipsychotics on it are developed. Finally, the last part concerns, initially, the recommendations of the American Psychiatric Association (APA), the World Federation of Societies of Biological Psychiatry (WFBP) and the National Agency for the Safety of Medicine (ANSM). In a second step, the possible role of the pharmacist in the accompaniment of these patients and the tools that he may have at his disposal are mentioned.
