Effets indésirables des z-drugs : analyse des cas notifiés par les médecins généralistes dans la base nationale de pharmacovigilance

Abstract : Introduction: zolpidem and zopiclone, also known as z-drugs, are indicated for the treatment of severe sleep disorders. The objective of this study is to determine the adverse effects (AEs) profile of the patients taking these drugs and to analyze the AEs according to their severity and then according to the gender. Material and method: the data come from a series of cases, corresponding to spontaneous declarations made by general practitioners registered in the national pharmacovigilance database. Results: from 1987 to 2018, 758 cases of AEs were reported, with 472 patients treated with zolpidem (62.3%) and 286 with zopiclone (37.7%). The statements concerned 284 men (37.5%) and 471 women (62.1%). The most common AEs were psychiatric and neurological conditions. 395 cases were considered severe (52.1%) and 363 were non-severe (47.9%). In the group of severe cases, neurological conditions were more numerous than in the group of non-severe cases (p = 0.01). We found more ocular AEs in women than in men (p = 0.04). Conclusion: our study shows that AEs with z-drugs can be numerous and sometimes severe. General practitioners should be encouraged to declare AEs more so that they are better known with these drugs.
