, Mon stage s'insère dans un axe de recherche qui consiste à modifier génétiquement la microalgue Phaeodactylum tricornutum. Il s'agit notamment de la modifier en surexprimant, sous-exprimant ou inactivant les gènes codant des facteurs de transcription (FTs) potentiellement impliqués dans la production de lipides. Le but de ce stage a été de : 1) sélectionner les FTs présentant une expression différentielle marquée en condition de stress azoté ; 2) construire les plasmides recombinés pour ces candidats en utilisant différentes méthodes de recombinaison génétique, Les microalgues sont des organismes d'intérêt pour la production de biocarburants car en condition de stress, elles produisent des triglycérides qui peuvent être transformés en biodiesel ou biokérosène

C. Auréochromes and . Hsf, Deux constructions plasmidiques pour les méthodes de sur et sousexpression ont été réalisées avec succès et la microalgue transformée par électroporation. Les résultats ne sont à ce jour pas connus. Les plasmides pour la méthode de KO ont été construits par « Golden Gate » et ont été utilisés dans la technique de transformation par conjugaison. Les résultats indiquent la présence de colonies de Pt résistantes à la zéocine sur les boîtes de Pétri, des transformations par électroporation et conjugaison bactérienne. L'analyse des données de séquençage ARN a fait ressortir 7 candidats appartenant à 5 familles
