Mise au point d'un ELISA pour la détection des auto-anticorps anti-interféron gamma

Abstract : Interferon gamma plays a key role in the fight against intracellular germs and nontuberculous mycobacteria (NTMs). NTMs are responsible for disseminated infections in patients producing neutralizing anti-interferon gamma autoantibodies. The detection of these autoantibodies is a diagnostic issue. In this work, we have developed an ELISA test for the detection of anti-interferon gamma antibodies in the blood of patients. Our ELISA test proved to be a repeatable and reproducible test. We determined a threshold of positivity from sera of healthy volunteers. To ensure the specificity of the antigen-antibody reaction we evaluated for each patient the signal obtained without antigen at the bottom of the well. We tested 162 blood samples selected on their clinical presentation (mainly M. tuberculosis infections) or on their ability to synthesize interferon gamma in the presence of a mitogen phytohemagglutinin. Six samples were found to be positive in ELISA. The prevalence of the anti-interferon gamma antibody in our population is therefore 3.8%. The passage of these samples in flow cytometry showed no inhibition of the phosphorylation of STAT1. The detected antibodies therefore do not have a neutralizing nature in this context. This work opens up new perspectives and our ELISA test could be an effective tool for the detection and characterization of anti-interferon gamma autoantibodies.
