Résumé, Introduction : L'immunoglobuline E (IgE) est le médiateur principal des maladies allergiques, dont les étiologies résultent d'interactions complexes entre des facteurs environnementaux, comportementaux et génétiques. Les maladies allergiques peuvent s'exprimer sous différentes formes, comme l'asthme. Dans l'asthme, l'association entre méthylation et IgE a déjà été retrouvée pour certains gènes, notamment SLC25A33

, L'objectif de ce travail était d'identifier les sites de méthylation associés aux taux d'IgE totales et spécifiques dans une population de sujets asthmatiques

, Méthodes : La méthylation, obtenue par méthylC capture-sequencing, a été analysée en fonction des taux d'IgE chez 357 adultes asthmatiques issus de la cohorte EGEA selon une approche par gène candidat (IgE totales et IgE spécifiques à Der p 1, 2, Fel d 1 et Phl p 1) et une approche sur l'épigénome entier

, Un nouveau site CpG (132009796) était associé aux taux d'IgE totales au sein du gène IL-4. L'analyse d'enrichissement montrait que les sites CpG ayant une valeur p (non corrigée) inférieure à 10 -4 étaient majoritairement présents au sein d'introns. Aucun site n'a été retrouvé associé aux IgE spécifiques. Conclusion : Ce travail a identifié de nouveaux sites de méthylation liées aux taux d'IgE totales, Résultat : L'approche sur l'épigénome entier a identifié 11 régions significativement associées aux taux d'IgE totales dont une concernant le gène IL-17Ra et une autre en amont de TUBGCP3
