Exploration in vitro de la réponse proliférative des lymphocytes T : mise en place de méthode et analyse de la réponse antigène spécifique

Abstract : T cells are major cells of the adaptive immune system whose coordination or cytotoxicity actions render them key players in the antigen specific response. Activation by the antigen induces lymphocyte proliferation or cytokine production. These proliferative and functional responses can be analyzed in vitro by the lymphocyte proliferation test (LPT). We implemented this technique at Caen University Hospital using a flow cytometry method by measuring the dilution of the CFSE. The development of this technique met two objectives. Firstly, to carry out quality controls of the apheresis bags used for patients treated with extracorporeal photochemotherapy (ECP) at Caen Etablissement Français du Sang. Our results validate the irradiation process by showing the decrease in the proliferation of T lymphocytes as well as the decrease in their post-ECP viability. Secondly, this technique can contribute to the diagnosis of delayed hypersensitivity. We explored a cow's amniotic fluid proteins contact dermatitis case and we were able to confirm the allergic mechanism for this patient.
