Validation d’un plan de suivi métrologique du pH en culture embryonnaire

Abstract : In a laboratory of medically assisted reproduction, the quality of embryonic development is closely linked to the environmental conditions to which embryos are exposed, notably in incubators. The pH of culture media is one of the essential critical parameters to be mastered in embryonic culture. The main goal of this study is the validation of the pH measurement method of our embryonic culture media. The secondary objectives are the evaluation of different conditions that can fluctuate the pH in the laboratory (handling of the media, incubation time of the media, presence of covering oil, CO2 content of the incubators). We chose to use a blood gas analyzer (RapidPoint 500) to perform the pH measurements. The culture media evaluated was continuous single culture complete (CSCM-C®) from IrvineScientific and the incubators tested were the K-Systems G210TM incubators. In order to validate this new method of pH measurement, pH stress tests followed by an evaluation of the performance of the method were carried out, in accordance with the requirements of COFRAC (French Committee for Accreditation). The performance tests carried out in this study showed good results and thus allow the validation of the method. The correlation study between the pH of the culture media and CO2 rate of the incubators allowed the definition of a new CO2 target value for the incubators in the laboratory: the new optimal rate is 7% with corresponding EMTs of +/- 1%. This new method of pH measurement of our environment will fit perfectly with our metrological strategy currently in place in the laboratory. Regular pH controls, established according to a frequency defined by the head biologist, will make it possible adapt the CO2 content of incubators and then will take part in the optimization of embryonic culture conditions.
