Cryobiologie appliquée aux leptopspires

Abstract : The issue of strain conservation is critical. A manufacturer must be able to ensure the homogeneity of production batches. Cultures of active growing bacteria are evolutionary. It is impossible to guarantee the immunogenic quality of the vaccine over time without starting from a primary and working homogeneous, viable, cultivable bank. Freezing also reduces the risk of contamination or loss of the cell line. The frozen bank is an asset, a real industrial heritage, to which we must be very vigilant. Premises, equipment and materials used are critical elements. Not all laboratories can invest in liquid nitrogen storage tanks with automatic filling, for example, nor in expensive programmable cryogenic freezers, for which qualifications are mandatory, time-consuming and costly. Here we discuss the two types of approaches, the industrial approach, which imposes complex specifications, equipment and qualified processes, but also the quality approach of a research and development laboratory that seeks to simplify administrative, technical and accounting procedures as much as possible on a daily basis and to devote itself to its profession, to innovation and to the development of veterinary vaccin. This is the case here with this leptospira canine vaccine. The disease evolves, so does the vaccine. New serogroups have to be frozen before being cultivated on a larger scale. It is essential to build up and secure the valuable stocks or cell banks of successful vaccine candidates. This study identifies critical parameters in the freezing of leptospira and exposes different models. From the programmable cryogenic freezer to Mr Frosty, from -80°C storage to nitrogen storage, here are some leads. No miracle recipe, no generalities, each working strain has its own story. The objective is to define a simple protocol that works for the greatest number of people. It is possible to make it simple, but above all it must be done seriously.
