Validation d’un antiseptique biodégradable : étude in vitro d’un extrait de Solidago virgaurea sur saccharomyces cerevisiae

Abstract : This work is part of the overall project to facilitate the Research and Development (R&D) of new "green", biodegradable active ingredients for oral hygiene products. Preliminary studies have shown that an extract from a plant, Solidago virgaurea (Bucovia ™) inhibits mixed bacterial-fungal biofilms formed with yeast, Candida albicans, and oral bacteria. This plant extract is by definition biodegradable. This work was a technical development, with a view to other bacteriofungal co-cultures for other applications of Bucovia, for example in dermatology for "green" cosmetics. We tested a mixed biofilm model with a yeast, Saccharomyces cerevisiae, and a bacteria representative of the oral biofilm, Streptococcus salivarius. We performed cultures in liquid medium and measured the surface area of S. cerevisiae cells with ImageJ imaging software. Bucovia did not inhibit the growth of S. cerevisiae, but it did decrease the size of the cells. In co-culture, Bucovia promoted the deposition of S. cerevisiae and S. salivarius at the bottom of the tubes and inhibited subcultures on agar media, even after direct exposure to Bucovia. The subcultures are an originality of this work. At the molecular level, Bucovia's mechanism of action remains to be elucidated. In conclusion, the destabilization of the biofilm and the inhibition of subcultures are two interesting properties of Bucovia. This active ingredient is promising in biodegradable "green" cosmetics, intended to control the proliferation of biofil.
