Données de survie et caractéristiques des patientes finistériennes traitées par inhibiteurs de PARP pour un cancer de l’ovaire entre 2014 et 2019 : étude rétrospective de l’efficacité et de la tolérance sur données du Finistère, en comparaison à celles de la littérature

Abstract : Ovarian cancer (OC) is the 4th leading cause of cancer death in women in France (National Cancer Institute 2019). The use of PARP inhibitors (PARPi) made it possible to improve the prognosis, as shown by the results of the SOLO2 study. No « real life » study had been carried out in Finistere on data from survival of patients treated for OC by PARPi. Methods : our study was retrospective, and multicentric, performed in the Finistere Area of Brittany in France. The primary endpoint was to assess the progression-free survival (PFS) of patients treated for relapsing platinum-sensitive OC by PARPi (olaparib or niraparib), between January 2014 and December 2019. Secondary endpoints were to assess overall survival (SG), safety, and predictive parameters of progression and prognosis. Results : 54 patients were included: 23 in the olaparib group and 31 in the niraparib group. PFS and OS were 16.8 and 37.8 months in the olaparib group and 5.4 and 20.4 months in the niraparib group, respectively. Major side effects were clinically, asthenia and nausea ; and biologically, anemia, neutropenia and thrombocytopenia. In multivariate analysis, being in the niraparib group and higher age were predictors of progression. Having a personal or family history of digestive cancer was considered an unfavorable prognostic factor. On the contrary, the presence of a mutation in the BRCA gene was a protective factor against progression and mortality. Conclusions : the « real life » data of our Finistere cohort are consistent with those of the literature. Other work will have to be carried out on a larger scale, in order to obtain the necessary hindsight and further improve the management of patients treated for OC.
