Stratégies galéniques actuelles dans le développement de formes ophtalmiques : application au syndrome de l'œil sec

Abstract : The eye is a very sensitive and complex organ of the human body. The cornea is in direct contact with the external environment and can be subjected to numerous attacks and pathologies. To treat them, many ophthalmic forms are developed and commercialized. However, due to various defense mechanisms and the complex anatomy of the eye, the passage of therapeutic molecules through the cornea is reduced. This results in a very low bioavailability of the API, requiring a high number of instillations to achieve therapeutic efficacy. Thus, the constant search for galenic improvements to overcome these problems is a real challenge. Several approaches exist and are presented in this thesis. Dry eye disease is a very common syndrome in the world population. It corresponds to a lack of tear fluid present on the surface of the cornea, which normally allows it to be nourished, hydrated and protected. This pathology is responsible for a permanent discomfort for the patient. It is therefore essential to treat this eye disorder by fully understanding its causes and mechanisms in order to adapt the treatment and have an effective cure. This report presents the development of an eye drop for the treatment of dry eye; the innovation is in the combination of two molecules never used together before. The objective is to demonstrate the synergy of this combination and its superior efficacy compared to other products already on the market.
