Applications nanobiothechnologies et thérapeutiques des membranes en bicouches lipidiques

Abstract : This thesis reviews the scientific literature and considers the ways to use biomimetic membranes for nanobiotechnological and therapeutic applications. About half of the drugs are targeted to membrane proteins incorporated into natural cellular bilayer membranes. We considered the therapeutic applications to include POC medicine, drug encapsulation and delivery, and implantable devices, either as the device itself or as a protective membrane for an implant. Lipid membrane-based biosensors for POC diagnostic represent the most important in vitro use of biomimetic membranes. Even so, proof of concept on biological samples is still lacking for these devices. Moreover, only a very few multidisciplinary teams are publishing on efficient POC testing devices that provide a rapid and accurate means for diagnostics. On the other hand, the main in vivo applications include drug delivery systems through liposome encapsulation. This field is already well documented. The building of artificial cells, biopiles or artificial organs and tissues is an emerging in vivo application of biomimetic lipid membranes. Another type of application is the use of lipid membranes for in vivo implantable biosensor devices, which would allow for the continuous real-time monitoring of specific biomarkers. As for artificial cells, the research in the field of in vivo biosensors is still at an early stage. Besides the discoveries and developments in the field of biomimetic lipid membranes for human medicine are easy to transfer to animal and environmental applications. This underlines the fact that veterinary and environmental applications are also considered for the ongoing development of this field of biomimetic lipid membrane research.
