Trouble déficitaire de l'attention avec hyperactivité‎ : état actuel des connaissances et projet de développement d'un agoniste 5-HT1A

Abstract : Hyperactivity is a common name for a pathology officially called Attention Deficit Hyperactivity Disorder (ADHD). The disease onset occurs during childhood, but remains in most cases in adults. Most of the treatments currently target the paediatric population, but there is an unmet medical need in adults, because of the lack of recognition of the disease in this population. Like any other psychiatric disorder, ADHD is complex in its diagnostic as well as in its treatment. The usefulness of pharmacological treatments has been shown many year ago. Psychostimulants are the reference treatment, but they induce toxicity problems. A non-psychostimulant molecule called Atomoxétine has been authorized in the recent years, but lacks in efficacy when compared to psychostimulants. Besides, it is not available in France and there is no indication in the adult population. Alternative treatments suffer from a lack of specificity and a high toxicity, and therefore there is a need for new medicines. The first part of this work will describe the disease and its diagnostic, and will review the existing treatments as well as the molecules currently in development. In the second part, the development program of a 5-HT1A agonist called osémozotan (MKC-242) will be proposed, the objective being to obtain a marketing authorization in ADHD both in adults and children.
