Développement d'un modèle original pour étudier la communication des cellules osseuses

Abstract : The healing potential of alveolar bone is sometimes insufficient to achieve ad integrum regeneration. To overcome this limitation, surgeons have to resort to bone regeneration strategies among them, tissue engineering appears as a solution. It applies the principles of engineering and life sciences in order to develop biologic substitutes. To improve cell viability and integration of the bone substitute, the fundamental understanding of the physiology, particularly focusing on existent cellular communication, is crucial. In this work, we evaluated the applicability of a specific cell medium to a unique tri-culture model, composed of human primary cells, namely osteoblasts, osteoclasts and endothelial progenitor cells. Our results show that this medium was able to maintain both osteoblastic and endothelial cell phenotype and obtain functional osteoclasts. These results allowed us to establish a tri-culture model. The validation of! this cell culture medium is the first step for the establishment of the conditions to study the communication between cells involved for bone regeneration. These studies will open new opportunities in the development of more efficient bone tissue engineering approaches.
