La galectine-9 comme biomarqueur d’activité au cours du lupus érythémateux systémique

Abstract : Introduction: the objective was to evaluate the performance of galectin-9 serum levels as biomarker for disease activity in a cohort of systemic lupus erythematosus patients. Methods: galectin-9 levels were measured with bead-based multiplex assay in a cohort of SLE patients who fulfilled ≥ 4 American College of Rheumatology classification criteria for the disease. As controls, healthy individuals matched for sex and age were included. Results: a total of 142 samples from 110 unique SLE patients were investigated. Serum levels of Galectin-9 were significantly higher in SLE patients as compared to controls (14.7±10.0 ng/mL vs 5.8±2.5 ng/mL). Levels were higher in patients with active disease, as defined by SLEDAI ≥ 6, than in patients in a quiescent state (19.7±12.0 ng/mL vs 11.1±6.4 ng/mL). The levels correlated significantly with “classical” activity biomarkers (SLEDAI scores, anti-dsDNA antibodies and complement C3 and C4 levels). Galectin-9 levels were greater in patients with arthritis, serositis, renal and haematological involvement as well as with anti-Sm/RNP antibodies. Conclusion: galectin-9 appears as a reliable biomarker for monitoring SLE disease activity. Particularly in systemic forms of the disease with serositis, renal and haematological involvement. Its performance to predict clinical involvement, to evaluate treatment response and for longitudinal follow-up should be further assessed in longitudinal cohorts of lupus patients.
