Analyse de la constitution chromosomique des zygotes mono- et poly- pronucléés par hybridation in situ par fluorescence et confrontation au nombre de pronuclei observés

Abstract : Objective: compare the pronuclear status of zygotes from ICSI with an abnormality of the pronucleus number to their chromosomal composition applied by fluorescence in situ hybridization (FISH), using probes specific for chromosomes X, Y and 18. Materials: cytological analysis using fluorescent probes for chromosomes X, Y and 18, of thirty-nine IVF-ICSI zygotes from 23 couples and with pronuclear numericals anomalies detected during the prior microscopic analysis. Results: 43% (3/7) of the monopronucleated zygotes were haploids and 23% (3/13) of the tripronucleated zygotes were triploids. 28.6% of the monopronucleated zygotes (2/7) and 15.4% (2/13) of the tripronucleated zygotes were diploid. Conclusion: there is a great difference between the evaluation of ploidy based on morphological parameters and the actual chromosomal composition of zygotes.
