Mise au point du dosage du 3-hydroxybenzo[a]pyrène urinaire et utilisation de ce métabolite comme biomarqueur d'exposition aux Hydrocarbures Aromatiques Polycycliques en population générale et professionnellement exposée

Abstract : Polycyclic Aromatic Hydrocarbons (PAH) are lung, bladder and skin carcinogens. Some industries and all the general population are exposed. Only benzo(a)pyrene is classified as carcinogenic to humans. However, biological monitoring of exposure is performed by 1-hydroxypyrene analysis which is a metabolite of pyrene, a non-carcinogen PAH. Thus, we developed a analytical method for quantification urinary 3-hydroxybenzo(a)pyrene (3OHBaP). Analysis begins with a solid-phase extraction step followed by chromatographic separation using HPLC and fluorescence detection. Detection limit was equal to 0.02 ng/L and precision and accuracy were validated. The method was applied to non- occupationally and occupationally subjects. The median concentrations of 3OHBaP in urine of smokers was equal to 0.023 nmol/mol creatinine, i.e. twice than in non-smokers. Levels were respectively 7 and 17 times higher among industrial workers of silicon and carbon electrodes. Finding levels of 3OHBaP in different sampling time have established that the optimal time of urine collection was at the post shift at the end of the week, when exposure was for a full week. Strong variability of regression equation from the correlation between urinary levels of 3OHBaP and 1OHP was observed between the two sectors. Thus, because composition of PAH mixtures depends on source, use of 3OHBaP in biological monitoring of PAH is essential.
