Recherche et validation de résistances génétiques au dépérissement bactérien causé par Pseudomonas syringae chez l'abricotier (Prunus armeniaca Linné)

Abstract : Bacterial canker of Prunus caused by Pseudomonas syringae is of major concern in apricot culture in France. The lack of chemical solution for this disease and the difficulty to predict damages lead us to consider the genetic improvement as a priority. However, little work has been done describing genetic resistance to P. syringae in stone fruits. Here, we studied resistance on leaves and branches of apricot tree (Prunus armeniaca L.) after inoculation of P. syringae pv. syringae on wounds (41A strain). Two biparental populations (pseudo-F1), 'Bergeron'×'Bakour' (224 individuals) and 'Goldrich'×'Moniqui' (196 individuals) and a collection of genetic resources have been selected for this work. This material was genotyped, by Single Sequence Repeat markers for biparental populations, and by Single Nucleotide Polymorphism for 76 accessions of the collection. Phenotypic data displayed a quantitative resistance, suggesting a polygenic model for this trait. A set of 40 SNPs showed association with resistance on branches and/or on leaves has been reported. Within the population 'Goldrich'×'Moniqui', linkage analysis showed a region of 4,8 to 8,8 cM (depending on the variable), at the end of the chromosome 4, accounting for 10,1 to 11,4% of the phenotypic variance on leaves. Associations to resistance on branches colocalised in this region. Two other regions on chromosomes 3 and 5 are also linked to resistance in both organs. Thank to the low extent of linkage disequilibrium and considering the available genetic and phenotypic diversity in apricot tree, we recommend to carry on with Genome Wide Association Studies (GWAS) for the genetic improvement in this species.
