Mise en place et développement d'une plateforme de culture pour la levure pichia pastoris

Abstract : Since the first marketing authorisation for a recombinant insulin in 1984, the share of biopharmaceuticals has been constantly growing up, reaching now 25% of the total pharmaceutical market. The rise of New Biological Entities has definitely led to changes in the health area : many laboratories are now investing in research and development to provide new therapeutic solutions for diseases such as cancer, diabetes or immune diseases. The development of antibody fragments, small molecules able to target less reachable epitopes compared to common antibodies, has increased the companies’ interest to use microorganisms as expression systems, because of the less complexity required. In this way, Merck Serono Biodevelopment at Martillac has recently extended its activities to the development and the production of recombinant molecules by microorganisms. In order to limit development process costs, complexity and duration linked to new clones implementation, the laboratory aims to set up development generic platforms. The objective of the experimental study has consisted in developing the fermentation step of the Pichia pastoris platform. First, a 24-well plate system has been implemented for fermentation miniaturization and high throughput screening purposes. The growth during batch process, and the evolution of biomass and the protein production from the overall process simulation have been evaluated. Once the system has been validated for growth studies, design of experiments have been shaped to develop a chemically defined medium with minimal compounds. Finally, screening and simulation data have been combined to evaluate P.pastoris ability to produce, in bioreactor, the molecule of interest with the previously developed minimal medium.
