Création et caractérisation de marqueurs SSR pour la fétuque rouge traçante - Festuca rubra subsp. Rubra - et étude de diversité génétique

Abstract : Turfgrass is a very important industry in the US. Awareness of the negative impacts of chemicals used for lawn maintenance as well as new customers and growers expectations, led to a new market: low-maintenance and low-input lawns. Because of its adaptation to poor, saline and dry environments, strong creeping red fescue, Festuca rubra subsp. rubra, perfectly matches low-maintenance requirements. This study aims at applying molecular techniques to this sub-species. 22 polymorphic SSR markers were tested on 95 strong creeping red fescue genotypes and then used on 37 fine fescue cultivars. AMOVA was realized on the binary matrix gathering the 610 obtained alleles. Neighbor Joining algorithm enabled us to build a phylogenetic tree via NTSYS software. Finally, bayesian algorithm was used to determine the structure and the most parsimonious number of populations via STRUCTURE software. Results revealed genetically identical cultivars and within-population variance of 76%, showing low germplasm diversity. Results obtained in this study can be used to differentiate fine fescues and help seed mixture designers choose the most efficient combination of varieties. Future research involves the use of fingerprinting for plant variety identification and protection.
