Validation fonctionnelle du rôle des miARNs dans la tolérance au phénanthrène chez Spartina : Utilisation d’Arabidopsis en système hétérologue

Abstract : The study of mechanisms by which plants tolerate environmental pollutants represents a major line of research in the prospect of environmental remediation. This internship project is based on the small-RNAseq data analysis of recently annotated MIR genes differentially expressed under PAH stress in Spartina (Poaceae, Chloridoideae). In order to confirm the function of differentially expressed miRNA families, we studied the effect of their mutation on Arabidopsis homologues. The validation of the sequencing data was performed by RT-qPCR and a set of candidate MIR genes was selected in order to identify mutant lines in Arabidopsis. A library of homozygous T-DNA mutant seeds corresponding to 31 lines (knock-out or knock-down mutants) of both MIR genes and targets of interest has been constituted. Phenotypic screening of phenanthrene effect over development of their primary root has shown phenotypes displaying hypersensitivity or tolerance, respectively associated to MIR156 and MIR159. The results of an inverse approach using miPEP treatments that specifically induce the overexpression of wild-type MIR genes seems to confirm these observations.
