Pertinence des méthodes vaccinales actuelles vis-à-vis de l'immunosénescence et nouvelles stratégies développées

Abstract : Vaccination is still one of the best protections that humans have designed to protect themselves from infectious diseases. The growing knowledge of the mechanisms regarding innate and adaptative immunities has enabled the development of a multitude of vaccines to address increasingly complex problems, while ensuring efficacy, safety and low cost. Despite all these advances, the ageing population characterized by immunosenescence brings new challenges to this vaccination. Indeed, this abstruse and continuous remodelling of the immune system is in addition to generalized aging, which is also characterized by genetic and functional alterations. The development of new vaccines is therefore confronted with growing and intereconnected issues. Far from being exhaustive, this thesis will focus on the relevance of our current vaccine methods with regard to immunosenescence, on the advent of systems biology as a new axis of vaccine research and development ; as well as on the advantages, defects and limitations of a vaccination that aim to be ever more personalized.
