Place des inhibiteurs de Janus Kinase dans la prise en charge de la polyarthrite rhumatoïde

Abstract : The rheumatoid arthritis is a chronic inflammatory rheumatism which affects more than 200 000 people in France. It is responsible for joint destruction that might be at the origin of fonctional disability and be source of difficulties in patient daily life. For 20 years, rheumatoid arthritis has known many scientific advancements. Those improvements about illness physiopathology have enable the discovery of new therapeutic targets. Recently a new class of medication has emerged about rhumatoid arthritis treatment: Janus Kinase inhibitors. JAK are intracellulars proteins kinase. Inhibitors will block signalling pathway and restrain the gene transcription encoding inflamatory proteins or proteins involved into autoimmune reaction. Currently, in order to treat RA, 3 JAK inhibitors have been approved as second-line therapy, many others are under development.
