Contribution à la mise au point d'un test de sensibilité variétale d'Actinidia sp. à Pseudomonas syringae pv. actinidiae

Abstract : Pseudomonas syringae pv. actinidiae (Psa), is the causal agent of the kiwifruit bacterial canker. Identified in Japan in 1984, it was first reported in Europe in 1994, in Italian orchards. Until recently, impact of bacterial canker on economy was relatively limited, but we are now seeing a significant spread of this bacterium and growth of damage. It has become a priority for the world kiwifruit industry, to improve our knowledge and control of this still relatively unknown Pseudomonas. Aim of the work carried out within the Ctifl, was to develop a test to investigate the varietal sensitivity of Actinidia sp . to Psa. In fact, obvious differences in susceptibility of kiwifruit varieties suggest that one solution to the problem of Psa can be found in choosing to grow new less susceptible varieties. Also , the season, early implementation of th e project and necessity to work in a confined laboratory, led us to develop a woody cane bioassay . Cane material is cut into lengths, wounded and Psa inoculum is placed into the freshly made wound. These canes are then incubated for 19 days under Psa conducive conditions. After this period of time, the bark on either side of the wound is cut longitudinally to expose and measure the length of the developing lesion beneath. The following day, the expression of bacterial ooze is recorded using a 0-4 scale. These two measurements are combined into a susceptibility index and this value can be used to compare the relative susceptibility of different Actinidia species and varieties to Psa infection. Despite encouraging results , we must remain vigilant to these study conclusions because some points remain to reconsider . Equally , the test still needs to be repeated several times to verify repeatability of obtained ranking lists.
